antigenic and molecular-weight characteristics which differ from those of CEA obtained from colon tumors (10, 13, 28). An assay specific for breast cancer-derived CEA would have potential usefulness for the early diagnosis and management of human breast carcinoma. However, highly purified extracts of breast CEA suitable for production of specific antisera or radiolabeling have not been prepared previously. With the ultimate goal of establishing a breast-specific CEA assay, we have partially purified CEA from human breast neoplasms. The recently described problems of aggregation and per chloric acid lability of CEA (31) and the relatively low concen trations of CEA in breast compared to colon carcinomata (5—8,

Immunological considerations in the use of goat antibodies to carcinoembryonic antigen for the radioimmunodetection of cancer.
Preparation of radioantibodies from carcinoembryonic antigen (CEA) antisera raised in goats by conventional procedures necessitates extensive absorption procedures to remove antibody activity against CEA-related substances. Further purification by affinity chromatography increases the antibody content of the radiolabeled preparation and enhances antibody uptake by a xenogeneic tumor. In patient...
متن کاملIsolation and purification of HLA-DR antigen from Daudi cell line by immunoaffinity chromatography
Introduction: The major histocompatibility complex (MHC) is a group of cell surface proteins that are essential for recognizing foreign molecules in human and other mammals. The physiologic function of MHC molecules is the presentation of peptides to T cells. In this study, we evaluated the purification of a class II MHC molecule (HLA-DR) from a human Burkitt′s lymphoma cell line; Daudi...
متن کاملPurification of human anti-erythropoietin polyclonal antibodies by precipitation and chromatography as an optimized method with potential application in vaccine studies
Introduction: Polyclonal antibodies are required to be affinity purified. Improved purification methods of polyclonal antibody provide an opportunity to pick the most purified immunoglobulins as a primary or secondary antibody in immunoassays that are included in many vaccine studies. Two common techniques for purifying proteins is salt precipitation and chromatography purification. Our work fo...
متن کاملThe isolation of carcinoembryonic antigen from tumor tissue at neutral pH.
Carcinoembryonic antigen (CEA) was purified from tumor tissue under mild conditions at neutral pH by a procedure that utilized affinity chromatography on concanvalin A. Further purification by gel filtration provided CEA in 10 to 20% yield and 10% purity. Antibody to this preparation was rendered specific for CEA by adsorption on a column of normal liver proteins bound to Sepharose. On reaction...
متن کاملMeasurement of Affinity Constant of Anti-human IgG Monoclonal Antibodies by an ELISA-based Method
Background: The affinity of an antibody to its antigen is a crucial parameter in its biological activity and performance of an immunoassay such as ELISA. Affinity of most IgG specific MAbs are often determined by methods which require labeling of either antigen or antibody, and are sometimes difficult to control, do not always lead to the expected signal and often result in immunological modifi...
